Ancestry.com takes 50% off its DNA testing services via coupon code "DNA50FTM2009". While not as big of a discount as our mention from last September, since expired, it's the best coupon we've seen on DNA testing so far this year. Shipping adds $4. Coupon expires March 1.

A best bet is the Paternal Lineage Test (Y-33) for $39.50 after the above coupon plus $4 for shipping. Note that this test is for male participants only.
